Canine Osteopontin (OPN) Quantitative Detection Kit [Kit Name] and Canine Type I Collagen (COL-1) Quantitative Detection Kit (ELISA) [Kit Use] are designed for the quantitative analysis of canine serum, plasma, and related liquid samples to determine the levels of Type I Collagen (COL-1). [Detection Principle] This kit utilizes a double-antibody two-step sandwich ELISA method. The plate is pre-coated with polyclonal antibodies specific to canine Type I Collagen (COL-1). Both the standard and the sample are added to the wells, allowing the target protein to bind to the immobilized antibody. After an incubation period, unbound materials are washed away. A labeled enzyme conjugate is then added, and the plate is incubated again to allow the enzyme-labeled antibody to bind to the captured COL-1. Following another wash step, substrates A and B are introduced sequentially. In the presence of horseradish peroxidase (HRP), the TMB substrate is converted into a blue product, which changes to yellow upon acidification. The intensity of the color is directly proportional to the concentration of COL-1 in the sample. The optical density (OD) is measured at 450 nm, and the concentration of Type I Collagen in the sample is calculated by comparing the OD values to a standard curve. This reliable and sensitive method ensures accurate quantification of COL-1 in various canine biological samples, making it a valuable tool for research and clinical diagnostics.
