This reagent is intended for research use only. The kit is designed to detect HIV-Gp41 in human serum, plasma, and related biological fluids. The experimental principle is based on the double-antibody sandwich enzyme-linked immunosorbent assay (ELISA). In this method, a solid-phase antibody, purified HIV-Gp41 antibody, is pre-coated onto microtiter plates. When the sample is added, it binds specifically to the immobilized antibody if HIV-Gp41 is present. Unbound substances are then washed away. Next, an HRP-labeled HIV-Gp41 antibody is introduced, which binds to the captured antigen, forming a complex of antibody-antigen-enzyme-labeled antibody. After thorough washing, the substrate TMB is added. Under the catalytic action of HRP, TMB turns blue and is then converted to yellow by the addition of an acidic stop solution. The optical density (OD) at 450 nm is measured using a microplate reader, and the result is compared against a pre-established cutoff value to determine the presence or absence of HIV-Gp41 in the sample.
The kit consists of several components, available in both 48-well and 96-well configurations. It includes a sealed bag with a coated plate, two sealing films, negative and positive controls, enzyme-labeled reagents, standard solutions, sample diluent, developer A and B, stop solution, and a concentrated wash solution. All components should be stored at 2–8°C. The negative and positive controls are provided in 0.5 mL volumes, while the enzyme standard reagent, sample diluent, developer A, and developer B are available in 3 mL or 6 mL quantities depending on the configuration. The concentrated wash solution comes in a 20 mL bottle, suitable for 20 or 30 dilutions, depending on the version. Proper storage and handling are essential to ensure accurate and reliable results.
